Objective To investigate effect of p27^kipl and cyclinE proteins on the genesis and progression ofhuman pancreatic cancer. Methods The expression of p27^kipl and cyclinE in tumor tissue and adjacent tissue of 32 patients with pancreatic cancer were detected by SP immunohistochemical technique. Results p27^kipl protein postitive-expression rate in tumor tissue of pancreatic cancer was 56%,which was lower than that in adjacent pancreatic tissue(P<0.05),p27^kipl protein positive-expression correlated significantly with tumor cell differentiation and lymphy node metastasis (P<0.05);cyclinE positive-expression rate was 69%, which was higher than that in adjacent pancreatic tissue(P<0.05),cyclinE positive-expression also correlated significantly with tumor cell differentiation and lymphy node metastasis (P<0.05).Conclusions p27^kipl and cyclinE proteins may play an important role in genesis and progression of pancreatic cancer.
Objective To clone,express and purify recombinant OPG/RANKL/TRANCE.Method RNA was extracted from the fetal liver and was reverse transcribed into cDNA.RANKL gene was amplified by RT-PCR with designed primers and cDNA as template. The product of PCR were cloned into His-tagged expression vector-pProEX^TM The and sent to sequence.Induced by IPTG, recombinant protein of the RANKL was expressed in Escherichia .coli stably. The protein was purified by Niion exchange colum. Result Weight of PCR product was 500 bp and sequence of it were correct. Molucular weight of recombinant protein was 20 kD.Conclusion ODF/OPGL/RANKL/TRANCE gene can be amplified simply and conveniently by RT-PCR from the cDNA of fetal liver. Recombinant RANKL protein could be obtained by inducing Prokaryotic expression of it.
Objectives To construct human BMP-2 and OPG coexpressing vector and determine its expression in mannalian cells in vitro. Methods Using the isolated total RNA from osteosacoma cell line MG63 as a template,the cDNA encoding hOPG was amplified by reverse transcription-polymerase chain reation (RT-PCR) method and cloned into mammalian expression vector pIRES2-EGFP endonucleases sites EcoR I and BamH I, and the BMP-2 encoding region gene was cloned into endonucleases site BstX I.Then the recombinant plasmid pIRES2-OPG-BMP2 was transformed into COS-7 cell line,OPG and BMP-2 expression were determined by Western blot assay.Results (1) The sequence of OPG cDNA obtained in this experiment was the same as that of reported, recombinant plasmid pIRES-BMP2-OPG was constructed successfully. (2) Human OPG and BMP-2 overexpression cell line COS-7 was selected and confirmed by Western blot analysis .Conclusion Human OPG encoding cDNA.OPG and BMP-2 coexpressing vector and cell lineage expressed OPG and BMP-2 were obtained. These datas suggested coexpressing human OPG and BMP-2 may be a promising and logical apporach for treatment of osteoporosis.
Objective To evaluate the limb and joint function after treatment the pelvic bone tumors by the microwave hyperthermia inserted antenna arrays .Methods 51 patients were treated by inserted microwave antenna arrays induced hyperthermia. All patient limb and joint functions were observed and analyzed from oncology,function and complications. Results There were 9 recurred cases and 7 of them died. Among the remaining 42 patients, 26 patients had excellent and good limb and joint function, 6 patients had fair function. Conclusions By the method of inserted microwave antenna arrays induced hyperthermia to treat the pelvic tumors the limb and joint function was excellent.
Objective To observe effects of meshed relaxing short incisions on the level of superoxide dismutase and malondialdehyde in rat tension skin flap, and to investigate the mechanism of meshed relaxing short incisions (MRSI) on wound healing process of tension skin flap. Method An experimental model was designed to investingate he changes of superoxide dismutase (SOD) and malondialdehyde (MDA) in rat shin flap tissue each period of wound healing (12h,24h,48h,72h after suture). In the meantime, the biochemical reaction were employed. Results Our results showed that SOD content in rat skin flap tissue from MRSI group were significant higher than that of hypertension group (P≤0.05), while MDA content was significantly lower (P≤0.05). Conclusion The decrease of MDA contents may be one of the causes that MRSI improves microcirculation of skin flap, and reduces edema and facilitates healing of skin flap.
Objective By hybridization in situ and biomechanical approach of platelet-derived growth fator mRNA(PDGFmRNA) and insulin-like growth factor mRNA(IGFmRNA), we discussed the influence of the platelet conentrated liquid on the healing of rabbit ulna fracture.Method We selected 24 New Zealand rabbits,divided them into 4 groups randomly (blank group,serum-control group,group with platelet concentrated liquid and group with bone graft and platelet concentrated liquid), and then made the fracture model on the middle of ulna which was fixed by finger armor plate.Before the operation, we drew out 6 ml blood from femoral artery, performed anti-coagulation with the Sodium Citrate and centrifugated by low and the followed high speed. We purified the white blood plate and injected it into the fracture position. The rabbits were killed at 1st,2nd,5th and 6th week.Qualitative analysis by hybridization in situ of PDGFmRNA and IGFmRNA and biomechanical measurement on the yth week sample were made. Result Bone callus could be seen on the radius spectimen in various degrees when the rabbits were killed at 1st. 2nd, 4th and 6th week, particularly in the last week. The average maximum destructive load on the fracture tip is higher to the control, and there is significant difference (P<0.01). Conclusion The local application of platelet concentration on the fracture tip can accelerate its healing.
Objective To evaluate the biomechanics of hollow-compression-screw in the osteoporotic femoral neck with composite calcium phosphate cement (CCPC). Methods Sixteen femurs of superior segment were randomly divided into two groups: augmentation group and non-augmentation group. CCPC was used in augmentation group. Result Augmentation with CCPC would improve the initial mobile force of hollow-compression-screw, the initial mobile force and the maximal axial pull-out strength for augmentation group,non-augmentation troup in creased from (192.7±14.0)N and (202.8±14.0)N to (328.5±34.7)N and (347.8±31.2)N. There was significant difference of two groups (P<0.01). Conclusion CCPC can enhance hollow-compression-screw fixation in osteoporotic femoral neck.
Objective To study the effect of sub-hypothermia on hypertension cerebral hemorrhage and content of serum endothelin (ET). Method 87 hypertension cerebral hemorrhage cases were divided into subhypothermia group and control group randomly, patients in subhypothermia group received subhypothermia therapy in NICU 6 hours after operation or hospitalization. We tested serum ET content with specific radioimmunoassay method 24 hours, 72 hours and 3 weeks after therapy, and compared them with control group. Result ET content of two groups increased apparently after 24 hours, ET of subhypothermia group was lower than that of control group(P<0.01); it recovered 3 weeks after therapy. Conclustion Subhypothermia techniques can apparently improve prognosis of hypertension cerebral hemorrhage patients, serum ET level may be one important index to evaluate severity of hypertension cerebral hemorrhage.
Compared with best supportive care,chemotherapy can prolong survival and improve quality of life, and symptoms associated with societry activities and disease. Chemotherapy can improve quality of life of patients with effective treatment and stable treatment. Treatment benefit was often underestimated using clinical relief rate, so which sould not chosen as index for evaluating effect of palliative treatment. Soo for patients with poor status, and objective of relieving symptoms, short term, low-dose chemotherapy should be suggested dure to its characteristic of low cost, short inhospitalization time, and favorable improvement of quality of life. For patients with good condition in limited advanced stage, radical treatment is objective, and hence single chemotherapy of standard dose or chemotherapy combining radiotherapy should be suggested.
