  • PURIFICATION AND SOME PROPERTIES OF CELLULASE FROM ODONTOTERMES FORMOSANUS (ISOPTERA: TERMITIDAE)
  • The purification of the cellulase from Odontotermes forrnosanus workers was achieved by using anion-exchange column of UNOsphere Q, BioLogic DuoFlow chromatography system. The purified cellulase was identified as an endoglucanase and some of its properties were investigated. The EGase activity was 807.5-fold as high as the initial enzyme activity using CMC as substrate and 14.4-fold using salicin as substrate. The enzyme preparations were homogeneous as judged by SDS-PAGE electrophoresis, molecular weight of which was 80 kDa and confirmed by 2-DE zymogram analysis. The enzyme was isoelectric at pH 6.4, which was active on CMC substrate.
  • EXPRESSION OF A BEE-VENOM PHOSPHOLIPASE A2 FROM APIS CERANA CERANA IN E,.qCHERICHIA COLI
  • The venomous phospholipase A2 (AcPLA2) coding reading region of the Chinese honeybee (Apis cerana cerana), which is composed of 405 bp encoding a mature glycosylated peptide with 134 amino residues was transformed into the expression vector pETblue-1. Then the recombinant vector was introduced into Escherichia coli Tuner (DE3) plac I for expression. Analysis result of SDS-PAGE showed that the expression products had a protein band of about 15 kD. Detection of western blot using ant-European honeybee (Apis mellifera) phospholipase A2 (AmPLA2) polyclonal serum as the first antibody showed that the expression products appeared a special blot same as the native AmPLA2.The result demonstrated that the AcPLA2 peptide had been expressed in E. coli and the AcPLA2 has the similar antigenicity as the AmPLA2.
  • BIOACTIVITIES AND MECHANISM OF SPIRO ENOL ETHER ANALOGUES AGAINST PIERIS RAPAE
  • Nineteen kinds of spiro enol ether analogues were screened with larvae of Pieris rapae for antifeedant activity. The results showed that the antifeedant activity of compounds No.20 and No.12 was higher than others. In non-choice test, AFC50 values within 24 h of compounds No.20 and No.12 against 3rd instar larvae of P. rapae were 226.93ug/mL and 370.00ug/mL, and that in choice test against 4th larvae were 280.54 ug/mL and 398.88 ug/mL, respectively. Compd. No.20 could prolong the eggs hatch time and reduce the haemolymph content and the protein content in haemolymph of 4th instar larvae obviously. Compd. No.20 could protect tested leaves and control larvae of P. rapae effectively.
  • CHANGES OF GLYCEROL CONTENT IN DIAPAUSE LARVAEOF THE ORANGE WHEAT BLOSSOM MIDGE, SITODIPLOSIS MOSELLANA (GEHIN) IN VARIOUS SEASONS
  • The glycerol contents in diapause larvae of the orange wheat blossom midge, Sitodiplosis mosellana (Gehin), collected from various seasons, were measured. The results showed that there was less glycerol content in larvae during living on the wheat head. Content of glycerol began to increase significantly when the larvae left the wheat head and entered the soil. A change trend of upper- lower- upper- lower in larvae glycerol contents during diapause in soil was observed from June to April of next year. More glycerol could be examined in larvae collected in summer and winter than in spring and autumn. There was not more glycerol in cocooned larvae than that in non-cocooned larvae during various seasons from the point of statistics. Comparing the glycerol content of larvae being diapause in the first year with that of larvae in the second year, there was yet no obvious difference when larvae were collected in the same season belonged to different years. Therefore, it is shown that the content of glycerol in larvae of the wheat midge in diapause is affected mainly by the seasons or diapause intensity.
  • REPELLENT AND OVIPOSITION DETERRENT ACTIVITIES OFTHE ESSENTIAL OIL FROM MIKANIA MICRANTHA AND ITS COMPOUNDS ON PLUTELLA XYLOSTELLA
  • Repellent and oviposition deterrent activities of the essential oil from Mikania micrantha and five volatile compounds including limonene, a-terpinene, linalool, B-caryophylene and verbenone on the diamondback moth (DBM), Plutella xylostella, was investigated in door and in net-house. The results showed that the essential oil of the M. micrantha had significant repellant effect (at flow 100-180 mL/min) and oviposition deterrent activity at dose 1020 uL/seedling for the DBM. In five volatile compounds, a-terpinene, limonene and linalool had significant effect on repellent and oviposition deterrent of the DBM moths, but verbenone and B-caryophylene, no significantly effect was observed in repellent and oviposition deterrent.
  • TEMPERATURE REQUIREMENTS FOR THE DEVELOPMENT AND SURVIVAL OF RICE STEMBORERS IN LABORATORY CONDITIONS
  • The effect of seven constant temperatures from 10 to 40℃ (10, 15, 20, 25 30, 35 and 40℃) on the development of eggs, larvae and pupae of rice stemborers viz., Chilo polychrysa (Meyrick), C. suppressalis (Walker), C. partellus (Swinhoe), Scirpophaga incertulas (Walker) ,S. innotata (Walker) and Sesamia inferens (Walker) were studied. The mean developmental period among constant temperatures (in days) of egg, larva and pupa of six borers differed significantly (P < 0.0001). The mean percent of development per day of egg, larva and pupa of all borers gradually increased with the increase of constant temperatures. The total developmental period was inversely decreased with the increase of constant temperatures. The lower threshold temperature was found between 10-15℃and higher threshold temperature between 35-40℃, where no development took place. The mean developmental zero was 8.57+1.71, 7.70+1.01, 8.56+3.25, 10.19+2.19, 8.64+2.68 and 7.91+0.82 for egg, larva and pupa of above-mentioned borers respectively. The total thermal constant of egg, larva and pupa was 705.56, 725.32, 703.30, 556.59, 655.34 and 837.95 degree- days for C. polychrysa, C. suppressalis, C. partellus, S. incertulas, S. innotata and S. inferens respectively. The degree- days required for oviposition of female moths of the six borers was calculated as 99.06, 90.85, 99.29, 75.16, 92.25 and 80.41 respectively. The total degree- days required completing a generation was 804.62, 816.17, 802.59, 631.75, 648.84 and 918.36 respectively.
  • SPECIES DIVERSITY AND COMMUNITY STRUCTURE OF SUCKING LICE IN YUNNAN, CHINA
  • On the basis of investigating 9 counties (towns) in Yunnan Province of China, the species diversity and community structure of sucking lice on the body surface of small mammal hosts are studied in the paper. Species richness (S) is used to stand for the species diversity. The calculation of community diversity index and evenness are based on Shannon-Wiener‘s method. 2745 small mammals captured from the investigated sites belong to 10 families, 25 genera and 41 species in 5 orders (Rodentia, Insectivora, Scandentia, Logomorpha and Carnivora) while 18165 individuals of sucking lice collected from the body surface of the small mammal hosts are identified into 4 families, 6 genera and 22 species. The species of sucking lice are much less than the species of their hosts. Most species of small mammals have their fixed sucking lice on their body surface. One species of small mammals usually have few species of sucking lice (1 to 4 species). The close species of the hosts in the taxonomy are found to have the same or similar dominant species of sucking lice on their body surface. The results reveal that the species diversity of sucking lice on small mammals is very low with a very simple community structure. The results also imply there may be a close co-evolution relationship between the lice and the hosts.
  • FOUR NEW SPECIES OF COENOSIA MEIGEN (DIPTERA: MUSCIDAE) FROM YUNNAN, CHINA
  • This paper describes four new species of Coenosia Meigen, 1826, namely C. angustifolia sp.nov., C. obscuriabdominis sp. nov. C. sparagmocerca sp. nov. and C. sponsa sp.nov. Type specimens are deposited in Institute of Entomology, Shenyang Normal University, Shenyang, China.
  • GENUS EI:IOTIS MEYRICK (LEPIDOPTERA: OECOPHORIDAE) NEW FOR CHINA
  • This paper reports the genus Erotis Meyrick for the first time from China. Two new species Erotis expansa sp. nov. and Erotis punctalosa sp. nov. are described. E. expansa sp. nov. is related to E. phosphora Meyrick both in appearance and in the male genitalia, but can be differentiated from the latter by uncus broader and slightly concave at middle of caudal margin, gnathos nearly as long as uncus and valva with costa almost straight in the male genitalia. E. punctalosa resembles E. expansa sp. nov. externally, but can be easily distinguished from the latter by the female genitalia: eighth tergite caudally almost straight; two signa large thorn-like, basal plate irregularly triangular in shape, with fine spines along anterior margin. The photographs of the adults and the genital structures of the new species are provided. A key to all the known species of Erotis is given.
